Sarandë and Vlorë are both towns on the sea in the south. Sarandë is smaller, faces Corfu, and is close to Ksamil and Butrint. Vlorë is bigger, has more going on all year, and is where the Riviera road starts.

Choose Sarandë if your trip is built around Butrint, Ksamil and the far south, or if you want the Corfu link. It is the better base for the southern coast and the worse base for everything else.
Choose Vlorë if you want a city that works in every season, a shorter transfer from Tirana, and the Llogara pass and Karaburun on your doorstep. It is less pretty and more useful.
If you are only visiting once and want the postcard coast, Sarandë wins. If you are considering living there, most people find Vlorë the easier place to spend a winter.
